چکیده

Metal ions are essential cofactors for functional expressions of many proteins in living organisms. In cells, several trace elements are needed to activate and stabilize enzymes, such as superoxide dismutase, metalloproteases, protein kinases, and transcriptional factors containing zinc finger proteins. Recently, intracellular metal trafficking proteins, that carry metal ions to specific target proteins, have been identified in various organisms including mammalians. Such proteins are thus called metallochaperones. This review focuses on the structural and functional characteristics of several metal transport systems, such as those of copper, iron, zinc, manganese, magnesium, and nickel.

A P-type ATPase importer that discriminates between essential and toxic transition metals.

Transition metals, although being essential cofactors in many physiological processes, are toxic at elevated concentrations. Among the membrane-embedded transport proteins that maintain appropriate intracellular levels of transition metals are ATP-driven pumps belonging to the P-type ATPase superfamily. These metal transporters may be differentiated according to their substrate specificities, w...

Physical interaction and functional coupling between ACDP4 and the intracellular ion chaperone COX11, an implication of the role of ACDP4 in essential metal ion transport and homeostasis

Divalent metal ions such as copper, manganese, and cobalt are essential for cell development, differentiation, function and survival. These essential metal ions are delivered into intracellular domains as cofactors for enzymes involved in neuropeptide and neurotransmitter synthesis, superoxide metabolism, and other biological functions in a target specific fashion. Altering the homeostasis of t...

Overexpression, purification, and site-directed spin labeling of the Nramp metal transporter from Mycobacterium leprae.

It has long been recognized that the pathogenicity of a broad range of intracellular parasites depends on the availability of transition metal ions, especially iron. Nramp1 (natural resistance-associated macrophage protein 1), a proton-coupled divalent metal ion transporter, has been identified as a controlling factor in the resistance or susceptibility to infection with a diverse range of intr...

Balance between metallothionein and metal response element binding transcription factor 1 is mediated by zinc ions (review).

Metal ion homeostasis and heavy metal detoxification systems are regulated by certain genes associated with metal ion transport. Metallothionein (MT) and metal response element binding transcription factor 1 (MTF‑1) are important regulatory proteins involved in the mediation of intracellular metal ion balance. Differences in the zinc‑binding affinities of the zinc fingers of MTF‑1 and the α‑ an...

Transition metal speciation in the cell: insights from the chemistry of metal ion receptors.

The essential transition metal ions are avidly accumulated by cells, yet they have two faces: They are put to use as required cofactors, but they also can catalyze cytotoxic reactions. Several families of proteins are emerging that control the activity of intracellular metal ions and help confine them to vital roles. These include integral transmembrane transporters, metalloregulatory sensors, ...
